| I. Background | |
| CD8 基因編碼的 CD8 分子是 T 細胞表面的關(guān)鍵協(xié)同受體，主要表達于細胞毒性 T 淋巴 細胞（CTLs）表面，由α（CD8α）和β（CD8β）兩個亞基通過二硫鍵連接形成異二聚體， 或以α-α同源二聚體形式存在。CD8 的核心功能是作為 T 細胞受體（TCR）的輔助分子，通 過其胞外段與主要組織相容性復(fù)合體 I 類分子（MHC I）結(jié)合，增強 TCR 對抗原-MHC 復(fù) 合物的識別敏感性，從而激活 T 細胞的細胞毒性功能，精準清除病毒感染細胞或腫瘤細胞。 CD8 基因位于人類 2 號染色體（2p12），包含多個外顯子，通過選擇性剪接產(chǎn)生不同 mRNA 亞型。其表達受轉(zhuǎn)錄因子 Runx3 等調(diào)控，在胸腺 T 細胞發(fā)育的陽性選擇階段起關(guān)鍵 作用，確保 T 細胞獲得識別自身 MHC 分子的能力。臨床中，CD8+ T 細胞是腫瘤免疫的核 心效應(yīng)細胞，其浸潤密度與多種癌癥（如黑色素瘤、結(jié)直腸癌）患者預(yù)后正相關(guān)，也是免 疫檢查點抑制劑（如抗 PD-1）的作用靶點。 |
