| I. Background | |
|
CLDN1 基因位于人類染色體 3q28.2，屬于緊密連接蛋白家族（Claudins）成員。CLDN1 基因編碼的緊密連接蛋白 Claudin-1 為四次跨膜蛋白，包含兩個細胞外環(huán)（ECL1、ECL2）和 N/C 端胞內尾。ECL1 通過二硫鍵穩(wěn)定結構，參與細胞旁通道調控；C 端與胞內支架蛋白結合，介導信號傳導。 在上皮細胞和內皮細胞中，Claudin-1 通過同型/異型相互作用形成緊密連接，調控細胞旁通透性。而 Claudin-1 與胞內蛋白（如 Zonula Occludens-1）結合，參與 Wnt/β-catenin、TGF-β等通路，影響細胞增殖、分化及遷移。在腫瘤中，其異常表達可激活促癌信號。
