				 抗體依賴的細胞介導(dǎo)的細胞毒性作用(ADCC,antibody-dependent cell-mediated cytotoxicity )是指抗體的 Fab 段結(jié)合病毒感染的細胞或腫瘤細胞的抗原表位,其 Fc 段與殺 傷細胞(NK 細胞、巨噬細胞等)表面的 FcR 結(jié)合,介導(dǎo)殺傷細胞直接殺傷靶細胞,它是 抗腫瘤的治療性抗體藥物發(fā)生作用的一種作用重要機制。 傳統(tǒng)用于測定 ADCC/ADCP 的方法主要依賴于相關(guān)原代免疫細胞的分離、體外分化, 進而測量靶細胞的殺傷或吞噬效應(yīng)。這些方法高度依賴供體原代細胞,價格昂貴且費時費 力,對原代細胞的分離分化過程對實驗操作的要求非常高,實驗失敗風(fēng)險較大,由于分離 分化細胞純度問題,還存在檢測信號低以及不穩(wěn)定均一的問題。另外,原代細胞具有不可 再生性,每次實驗的細胞供體來源可能都不一致,實驗結(jié)果可重復(fù)性較差,可能存在批次 差異。由于以上這些因素的影響,在需要高質(zhì)量控制的藥物開發(fā)過程中,很難建立穩(wěn)定可 靠的檢測方法。  |
