| I. Background | |
| 促紅細胞生成素（EPO）是一種由腎臟（成人）或肝臟（胎兒）分泌的糖蛋白激素， 主要功能是調(diào)控紅細胞的生成。EPO 屬于Ⅰ型細胞因子超家族，由 165 個氨基酸組成 4 個α 螺旋。在人類中，由于糖基化水平高，腎產(chǎn)生的 EPO 在血漿中的半衰期為 5-6 小時。當紅 細胞水平下降時，腎小管間質(zhì)細胞檢測到相對缺氧，并以經(jīng)典的內(nèi)分泌方式將 EPO 分泌到 循環(huán)中。然后，EPO 遷移到骨髓，在紅系祖細胞上結(jié)合同二聚體 EPO 受體（EPOR）2，促 進紅細胞生成。 EPOR 屬于 I 型細胞因子受體超家族，為單次跨膜蛋白，由胞外配體結(jié)合域、跨膜域及 胞內(nèi)信號域組成。EPO 結(jié)合 EPOR 后誘導(dǎo)受體二聚化，引發(fā)胞內(nèi) JAK2 自磷酸化，激活下 游增殖、分化及抗凋亡信號通路。 |
