| I. Background | |

OX40(也稱為 ACT35、CD134 或 TNFRSF4)屬于 TNFR 超家族，是一種Ⅰ型跨膜蛋白。OX40 主要在活化的 CD4+ T 細胞和 CD8+ T 細胞上表達，而在 NK 細胞和 NKT 細胞上表達水平相對較低。OX40L(也稱為 CD252、TNFSF4、CD134L 或 GP34)是 OX40 的配體，是一種 II 型糖蛋白。OX40L 主要表達在抗原呈遞細胞上，如 B 細胞和樹突狀細胞。
與 TNF 家族的其他成員類似，OX40 信號通過 TNF 受體相關因子傳導。OX40 信號在體內(nèi)通過 TRAF2 和 TRAF5 以及在體外通過 TRAF1、TRAF3 和 TRAF5 轉導到 T 細胞。OX40 和 OX40L 的表達受多種因素的調控。OX40 在 T 細胞上通過 TCR、CD28/CD80、CD40/CD40L 等信號誘導表達，在 T 細胞活化后 48-72 小時達到表達高峰。TCR 信號可以啟動 OX40 在多種細胞上的表達，而 CD28 和其他細胞因子可以進一步促進其在活化 T 細胞上的表達。IL-2、IL-4 和 TNF 可增強或延長 OX40 的表達。OX40/OX40L 在增強效應 T細胞功能、維持效應 T 細胞存活、抑制效應 T 細胞凋亡等方面發(fā)揮著關鍵作用。 |
